摘要:
通过对3种抗生素的筛选,确立最适抑制农杆菌过度生长、促进微型月季愈伤组织生长与分化的抗生素及其浓度,从而建立稳定而有效的微型月季遗传转化体系。结果表明:微型月季叶片培养于150 mg/L头孢噻肟钠培养基可抑制农杆菌生长且利于材料生长分化。侵染最佳条件为:侵染菌液OD600为0.5—0.6,侵染5—10 min,共培养2 d。‘荷仙姑’‘彩虹’和‘笑脸’微型月季阳性愈伤的PCR阳性率分别为26.4%,18.4%和31.2%。该研究结果为微型月季农杆菌遗传转化的抗生素选择和培育条件的优化提供一定参考。
Abstract:
By using three antibiotics (cefotaxime sodium,carbenicillin and ampicillin),the most suitable antibiotics and their concentrations for inhibiting excessive growth of Agrobacterium and promoting the growth and differentiation of miniature rose leaf callus were determined, thus to establish a stable and effective genetic transformation system for miniature roses. The results indicated that the transforming leaves cultured in 150 mg/L cefotaxime sodium medium could inhibit the growth of Agrobacterium and promote callus growth and differentiation. The optimal conditions for infection were as follows: an OD600 value of 0.5—0.6 in the infection solution, infection duration of 5—10 min, and co-cultivation for 2 days. The PCR positive transformants rates of ‘Hexiangu’ ‘Rainbow’ and ‘Smiling Face’ miniature rose positive callus were 26.4%, 18.4%, and 31.2%, respectively. The results could provide some reference for the selection of antibiotics and optimization of cultivation conditions in the genetic transformation of Agrobacterium.
